High-performance liquid chromatography (HPLC) is a technique used to separate, identify, and quantify each component in a mixture. HPLC relies on pumps to pass a pressurized liquid solvent containing the sample mixture through a column filled with a solid adsorbent material. Each component in the sample interacts slightly differently with the adsorbent material, causing different retention times for the components and leading to the separation of the components as they flow out of the column. Operating parameters such as mobile phase composition, stationary phase chemistry, flow rate, and temperature provide a wide range of separation applications. Maximum operating pressures up to around 4500 psi are common during HPLC testing (dependent upon column chemistry).
Ultra-High performance liquid chromatography (UPLC) uses particle size smaller than those used in HPLC applications. Maximum operating pressures of UPLC up to 15,000 psi provides better separations and faster analysis times. The most common is UV detection but others include Refractive Index (RI), Conductivity, Fluorescence and Mass Spectrometry.
